L-Theanine and GABA: Do They Work the Same Way?

Absorption and Brain Entry Are Two Separate Questions

L-theanine is absorbed in its original form, and its time course in humans has been well documented. When volunteers were given a dose of 25 to 100 mg, their plasma concentrations reached a peak after about 50 minutes. Its absorption and elimination half-lives are approximately 15 and 65 minutes, respectively. However, that study did not measure brain concentrations, and its authors regard entry into the brain as an assumption based on data from rats.1

The case of GABA is more difficult because the molecule is small but carries an electric charge at physiological pH, which hinders its ability to passively cross membranes. A review of GABA as a food supplement stated that the human evidence regarding its passage through the blood-brain barrier is contradictory rather than conclusive. It raised the enteric nervous system as an alternative. The review also mentioned a possible conflict of interest in a great deal of the human evidence that is in support of GABA.2

There are few direct measurements on either side, so the asymmetry lies upstream. Theanine is a neutral amino acid, a class that the barrier transports. GABA, on the other hand, is charged. That argues against the same route, which is why its case has remained open for decades.

How Strong Is the Case for Pairing Them?

The foundation is rodent work. In a 2019 study, mice and rats received a GABA and theanine mixture at 100 and 20 mg per kilogram of body weight. As a result, sleep latency decreased, while sleep duration increased compared with the effect of each substance individually. These effects were also accompanied by changes in EEG activity during both REM and non-REM sleep.3 Two points are important. First, sleep latency was measured after pentobarbital induction. Second, part of the EEG analysis used a caffeine-arousal model. Therefore, these tests should be regarded as pharmacological screenings rather than studies of natural sleep.

Human data were obtained in 2026. In the study, nineteen adults took 700 mg of GABA and 200 mg of theanine for a period of four weeks, after which their mean PSQI scores improved from 9.4 to 6.3, where a lower score means better sleep.4 The study was single-arm and included no placebo group nor any comparison with either GABA or theanine alone, so it only shows a signal and not a synergy.

Beyond the Purity Number

A figure obtained from an assay gives no information regarding stereochemistry. Although tea contains the L form, certain synthetic methods produce a mixture of both D and L forms, a mixture which a purity figure will not be able to separate. Optical rotation does provide such information. The two forms cause light to rotate in opposite directions, so a racemic mixture gives a reading near zero, whereas a value inside a narrow positive range shows that the material is mainly L. This number appears on the certificate for the bulk ingredient, upstream of the capsule.

The Takeaway

Theanine’s entry into the bloodstream has been measured. It is not known whether an oral dose of GABA gets to the brain. Neither of these facts decides which of the two is superior, and confusion arises when they are regarded as being the same. For someone considering the two options, the more comprehensive human record is that of theanine.
